چکیده

Received 2 January 2003; accepted 29 January 2003 ABSTRACT The tracing of neural pathways continues to be an important concern for neuroanatomy. During the second half of the nineteenth century, at a time when the foundations of neuroanatomy were being established, tracing pathways often required laborious dissections by hand. Then, neuroanatomy experienced a methodological revolution in the beginning of the 1970s with the development of powerful techniques based on the axonal transport of tracers. Axonal tracing of neuronal pathways using anterograde and retrograde transport is available. Axonal transport can be combined with immunohistochemistry for the neurochemical characterization of specific neuronal pathways. After tracing, tissue is collected following appropriate survival time for the tracer to be transported. Immunohistochemical methods, virus immunolabeling or autoradiography at the axon terminals are used for expressing the tracers. Retrograde axonal transport is used by certain viruses (e.g., herpes and pseudorabies virus) to spread from one neuron to the next in a chain of neurons (transneuronal transfer). It is also the method whereby toxins (e.g., tetanus) are transported from the periphery into the central nervous system. This has led to the idea that viral tracing may be an important way as a possible gene delivery method for the gene therapy in nervous system diseases.

Neuroanatomical tracing of neuronal projections with Fluoro-Gold.

The study of neuronal connectivity requires the ability to trace axons from the neuronal cell body to its axon terminal (anterograde tracing) and from the terminal back to the soma (retrograde tracing). Such neuroanatomical tracing is frequently used to identify neurons on the basis of their pre- or postsynaptic connections. Neuroanatomical tracing has become particularly important in nervous s...

NeuriteTracer: a novel ImageJ plugin for automated quantification of neurite outgrowth.

In vitro assays to measure neuronal growth are a fundamental tool used by many neurobiologists studying neuronal development and regeneration. The quantification of these assays requires accurate measurements of neurite length and neuronal cell numbers in neuronal cultures. Generally, these measurements are obtained through labor-intensive manual or semi-manual tracing of images. To automate th...

Multiple Structure Tracing in 3D Electron Micrographs

Automatic interpretation of Transmission Electron Micrograph (TEM) volumes is central to advancing current understanding of neural circuitry. In the context of TEM image analysis, tracing 3D neuronal structures is a significant problem. This work proposes a new model using the conditional random field (CRF) framework with higher order potentials for tracing multiple neuronal structures in 3D. T...

Evaluation of recombinant adenovirus-mediated gene delivery for expression of tracer genes in catecholaminergic neurons

Selective labeling of small populations of neurons of a given phenotype for conventional neuronal tracing is difficult because tracers can be taken up by all neurons at the injection site, resulting in nonspecific labeling of unrelated pathways. To overcome these problems, genetic approaches have been developed that introduce tracer proteins as transgenes under the control of cell-type-specific...

Magnetic Resonance Imaging of Neuronal Connections in the Macaque Monkey

Recently, an MRI-detectable, neuronal tract-tracing method in living animals was introduced that exploits the anterograde transport of manganese (Mn2+). We present the results of experiments simultaneously tracing manganese chloride and wheat germ agglutinin conjugated to horseradish peroxidase (WGA-HRP) to evaluate the specificity of the former by tracing the neuronal connections of the basal ...
